Pemeriksaan RPR dan Widal

TES RPR

     Tes RPR (Rapid Plasma Reagin) adalah suatu tes untuk mengetahui ada atau tidaknya antibodi terhadap kuman Treponema Pallidum.Tetapi tes ini menggunakan antigen yang non spesifik (dengan kata lain ni, antigen ini dibuat semirip mungkin dengan antigen Treponema).Tapi ada juga kok pemeriksaan Treponema dengan menggunakan antigen yang  berasal dari kuman Treponema(antigen spesifik),tapi antigen ini haraganya lebih mahal, pemeriksaan ini disebut dengan pemeriksaan TPHA.Kuman Treponema ini menyebabkan penyakit yang disebut Syphilis(Raja Singa).Penyakit Syphilis ini menular hlo,,terutama kalau  seorang penderita Syphilis melakukan kontak seksual dengan orang lain,pasti orang ini akan tertular penyakit ini.
Cara melakukan tes:
Alat dan Bahan
1.Sampel serum
2.Mikro pipet 50 ul
3.Rotator 
4.Antigen RPR siap pakai
5.NaCl 0,9%
6.Lempeng penguji
7.Lidi
Cara Kerja
  KUALITATIF
    1.Pipet 50 ul sampel,letakkan pada lempeng penguji
    2.Tambah 1 tetes reagen RPR (Carbon Antigen)
    3.Campur dengan lidi
    4.Goyang di Rotator selama 8 menit dengan kecepatan 100 rpm
       Baca Hasil:
      (+) terjadi flokulasi (endapan/presipitat) warna hitam
      (-)tidak terjadi flokulasi (endapan/presipitat) warna hitam

           Sebenarnya dalam tes ini masih ada lanjutan pemeriksaannya kalau tes kualitatifnya hasilnya positif,yaitu dilanjutkan dengan tes kuantitatif,dan karena tes kuantitatif ini sangat rumit cara kerjanya,,jadi saya tidak akan mempostingnya,hehehehehe




TES WIDAL
 Tes ini digunakan untuk mengetahui ada atau tidaknya antibodi terhadap kuman Salmonella dalam darah.Pemeriksaan ini dilakukan setelah 1 minggu diduga terinfeksi kuman Salmonella.Kuman Salmonella ini menyebabkan penyakit Demam Typhoid(typhus).Tanda-tanda penyakit typhus:
1.lidah kotor
2.leukopeni
3.limfositosis relatif
4.ujung lidah tremor
 Penderita yang mengalami infeksi kuman Salmonella maka sistem imunologinya menghasilkan antibodi yang yang menimbulkan aglutinasi yang bila kemudian dicampur dengan suspensi penyebab penyakit tersebut.


Cara Kerja:
Alat dan Bahan:
1.sampel serum
2.pipet tetes
3.tusuk gigi
4.antigen Salmonella typhi O
5.antigen Salmonella typhi H
 6.antigen Salmonella paratyphi BH
 7.antigen Salmonella paratyphi BO.
8.Objek glass


Prosedur:
1.Dengan pipet 0,2 ml letakkan 0,08 ml serum di atas 4 buah objek glass
2.Tambah 1 tetes
   antigen Salmonella typhi O,antigen Salmonella typhi ,.antigen Salmonella paratyphi BH,antigen 
   Salmonella paratyphi BO, diatas objek glass yang telah diberi serum dengan jarak 1 cm tegak lurus
3.Campur hingga homogen
4.Putar diatas meja selama 1 menit,
    Baca Hasil:
    (+)terjadi aglutinasi
    (-)tidak terjadi aglutinasi




Dalam tes ini juga masih ada pemeriksaan lanjutan jika hasil tes kualitatif positif,yaitu dengan tes kuantitatif ,,tetapi jika ada yang pengen tahu caranya,,
